Nucleic acid reactivity and conformation. II. Reaction of cytosine and uracil with sodium bisulfite.

چکیده
Treatment of calf thymus DNA with sodium bisulfite gave no deamination of cytosine to uracil, under conditions where 68 % of the cytosines of heat-denatured DNA were converted to uracil. This implies that the mutagenic properties of bisulfite are due to reaction with single-stranded DNA. Similarly, poly(1) . poly(C) was unreactive to bisulfite under conditions where poly(C) reacted readily. It was established by ultraviolet and circular dichroism studies that the initial step of the deamination sequence, the addition of bisulfite to the 5-6 double bond of cytosine, was inhibited in double-helical polynucleotides. In accord with this, it was found that, at 25”, bisulfite did not react with the uracils of poly(A) .poly(U) and poly(A) .2 poly(U), under conditions where it added to the 5-6 double bond of the uracils of free poly(U). At higher temperatures below the melting temperature, a slow reaction of poly(A) .2 poly(U) was observed. A number of factors may contribute to the resistance of multiple-helical polynucleotides to reaction with bisulfite. These include the inability of the modified base to take part in Watson-Crick hydrogen bonding and steric hindrance to the approach of the reagent in a multiple helix.

Effect of sodium bisulfite modification on the arginine acceptance of E. coli tRNA Arg.
Escherichia coli tRNA Arg was treated with sodium bisulfite to convert exposed cytosine residues to uracil. This treatment resulted in the loss of amino acid acceptance of the tRNA Arg with pseudo first-order reaction kinetics. The active and inactive molecules were separated after about 60e active and inactive molecules were separated after about 60 percent inactivation and analyzed for U in v...
متن کاملProfiling DNA Methylation Using Bisulfite Sequencing (BS-Seq).
DNA cytosine methylation is one of the most abundant epigenetic marks found in the plant nuclear genome. Bisulfite sequencing (BS-Seq) is the method of choice for profiling DNA cytosine methylation genome-wide at a single nucleotide resolution. The basis of this technique is that the unmethylated cytosine can be deaminated to uracil by sodium bisulfite, while the methylated cytosine is resistan...
متن کاملControl of carry-over contamination for PCR-based DNA methylation quantification using bisulfite treated DNA
In this study, we adapted the well known uracil DNA glycosylase (UNG) carry-over prevention system for PCR, and applied it to the analysis of DNA methylation based on sodium bisulfite conversion. As sodium bisulfite treatment converts unmethylated cytosine bases into uracil residues, bisulfite treated DNA is sensitive to UNG treatment. Therefore, UNG cannot be used for carry-over prevention of ...
متن کاملAccurate sodium bisulfite sequencing in plants.
DNA cytosine methylation is a conserved epigenetic modification frequently correlating with transcriptional silencing in a wide variety of eukaryotic organisms. Sodium bisulfite treatment of DNA converts unmethylated cytosine to uracil, while 5-methylated cytosine is protected. We describe techniques that ensure reliable sequencing data following sodium bisulfite conversion and to avoid common ...
متن کاملRecognition of a cytosine base lesion by a human damage-specific DNA binding protein
Sodium bisulfite reacts with cytosine and 5-methylcytosine, forming the 5,6-dihydrosulfonate adducts which deaminate to the uracil and thymine adducts, respectively. At alkaline pH, the sulfonate groups are then released, generating uracil and thymine. In DNA, the resulting G:U and G:T base mismatches generated are potential sites of mutagenesis. Using a human damage-specific DNA binding protei...
